Changes in Collagen Structure in Bone and Tail Tendon From Diabetic Rats Revealed By Atomic Force Microscopy

چکیده
INTRODUCTION Type 2 Diabetes (non-insulin-dependent diabetes mellitus, T2D) has detrimental impacts on multiple organ systems including the renal, circulatory and nervous systems. Destructive effects are also apparent on the musculoskeletal system, although studies are limited in comparison. T2D likely affects bone and other collagen-based tissues through multiple mechanisms. Chronic hyperglycemia in T2D has been implicated in the formation of advanced glycation end products (AGEs) which form when reducing sugars react with free amino groups in proteins. The formation of AGEs in Type I collagen could lead to a stiffening of fibrils and, in the case of bone, this may be lead to decreased toughness. However, few studies have directly investigated the collagen ultrastructure in diabetic tissues. The purpose of this work was to study the nanoscale morphology of Type I collagen in mineralized and non-mineralized tissue from normal and diabetic rats.

Nerve collagens from diabetic and nondiabetic Sprague-Dawley and biobreeding rats: an atomic force microscopy study.
BACKGROUND Alterations in rat's nerve collagens due to diabetes may be related to the permanence of damage due to diabetic neuropathy. We (1) provide a methodology for determining the diameters of collagen fibers accounting for atomic force microscope (AFM) imaging artifacts, (2) present data on structural differences in sciatic nerve endoneurial, epineurial and tail tendon collagens of control...
متن کاملNanoscale changes in collagen are reflected in physical and mechanical properties of bone at the microscale in diabetic rats.
Diabetes detrimentally affects the musculoskeletal system by stiffening the collagen matrix due to increased advanced glycation end products (AGEs). In this study, tibiae and tendon from Zucker diabetic Sprague-Dawley (ZDSD) rats were compared to Sprague-Dawley derived controls (CD) using Atomic Force Microscopy. ZDSD and CD tibiae were compared using Raman Spectroscopy and Reference Point Inde...

PURPOSE/AIM Collagen's role in bone is often considered secondary. As increased attention is paid to collagen, understanding the impact of tissue preservation is important in interpreting experimental results. The goal of this study was to test the hypothesis that bone fixation prior to demineralization would maintain its collagen ultrastructure in an undisturbed state when analyzed using Atomi...
متن کاملExperimental Diabetes Alters the Morphology and Nano-Structure of the Achilles Tendon
Although of several studies that associate chronic hyperglycemia with tendinopathy, the connection between morphometric changes as witnessed by magnetic resonance (MR) images, nanostructural changes, and inflammatory markers have not yet been fully established. Therefore, the present study has as a hypothesis that the Achilles tendons of rats with diabetes mellitus (DM) exhibit structural chang...
متن کاملInvestigations into the polymorphism of rat tail tendon fibrils using atomic force microscopy.
Collagen type I displays a typical banding periodicity of 67 nm when visualized by atomic force or transmission electron microscopy imaging. We have investigated collagen fibers extracted from rat tail tendons using atomic force microscopy, under different ionic and pH conditions. The majority of the fibers reproduce the typical wavy structure with 67 nm spacing and a height difference between ...
